The Inhibition of CD40/CD40L Expression by Buyang Huanwu Decoction in Rats with Myocardial Ischemia

【摘要】:正Aims In recent years,inflammation has been recognized as a major driving force in ischemic process, and increasing evidence has shown that increased levels of inflammatory markers,including C-reactive protein,NF-kappa B and matrix metalloproteinases,are related to Ischemic heart disease(IHD).BuYang HuanWu Decoction (BYHWD) is a well-known Chinese medicine formula.Some studies have recently reported that BYHWD can be used to treat ischemic coronary heart disease;little is known about the mechanism by which BYHWD alleviates ischemic heart disease(IHD).This study investigatethe potential mechanism by which BYHWD protects myocardial tissues from ischemia.Main methods A rat model with isoproterenal(ISO)-induced myocardial ischemia was established. Different doses of BYHWD(25.68,12.84 and 6.42 g·kg~(-1)) were lavaged to these rats,respectively. Then the superoxide dismutase(SOD) activity,the malondialdehyde(MDA) content and the free fatty acids (FFA) content in the serum were measured by biochemistry.The expression of the cluster of differentiation 40 (CD40) in the whole blood was measured using flow cytometry and the expression of CD40 and its ligand (CD40L) in myocardial tissues were determined by Western blotting.Key findings The results show that the SOD activities of the BYHWD pre-treated groups(25.68,12.84 and 6.42 g·kg~(-1)) were significantly higher than that of the model group,while the MDA and FFA contents of all the BYHWD pre-treated groups were less than that of the model group.Pre-treatment with BYHWD could decrease the expression of CD40 in the serum and the CD40 and CD40L expression in myocardial tissues. Significance The present study shows that BYHWD has a protective effect against ISO-induced myocardial ischemia and this effect may be attributed to the down-regulation of CD40 and CD40L expressions.Moreover,BYHWD can elevate the SOD activity,decrease the MDA level and FFA content in the serum,which suggests that the protective effect of BYHWD is related to these two actions as well.Our results provide an explanation for the relationship between the anti-inflammatory,protective effect of BYHWD on myocardial ischemia,and therefore,are valuable in understanding the mechanism of cardiovascular protection by BYHWD.The findings of the present study may shed light on the pharmacological basis for the clinical application of the traditional Chinese medicine in the treatment of ischemic cardiovascular diseases.
