Different Effects of low-dose hydroxyurea treatment on human ovarian cancer cells UACC-1598 sub-clones containing different double minutes

【摘要】:正Gene amplification is the most frequent aberrance in the tumorigenesis and progression of various tumors,and double minutes(dmin) is one of the identifiable gene amplification cytogenetically. A lot of proto-oncogenes and chemo-resistance genes are localized on extrachromosomal dmin,it offers advantage for tumors growth and resistance to chemotherapy.In this manuscript,we first established several sub-clone cells with apparent difference in the number of dmin by infinite dilution. Two sub-clone cells,one of which had generous dmin(the Number 1) and the other had scanty dmin(the Number 8),were chosen to do the further assay.Low-dose hydroxyurea were used to induce the elimination of dmin.Then we certified the evident difference between two sub-clone cells in proliferation,colony formation and migration.The results show that there are significantly different biological phenotypes in the two sub-clone cells containing different dmin.And different inhibitory effects were obtained after low-dose hydroxyurea treatment.It suggests that we should take different prescription to aim directly at neoplastic patient with dmin or not when hydroxyurea was used to treat tumor in clinic.
