Gene expression profile in cardiomyocyte-specific Smad4 knock-out mice

【摘要】:正We have demonstrated that cardiac-specific knock out Smad4 resulted in cardiac hypertrophy and heart failure.The results reveal an important function of Smad4 in cardiac remodeling,and suggest that an altered cellular response to TGF-#61538;could be a mechanism by which cardiac myocytes undergo hypertrophy.To further elucidate the molecular mechanism of cardiac hypertrophy caused by Smad4 deletion,We utilized cDNA microarrays to screen the altered gene expression in Smad4 knock-out mice.Total RNA extracted from the left ventricles from Smad4 mutant(n=5)and wild-type(n=5)mice was hybridized to Mouse Genome 430 2.0 Array of Affymetrix.Comparison of RNA expression levels in Smad4 mutant to wild-type myocardium revealed that about 103 genes were differentially expressed.The results demonstrated that genes related to the cardiac structure,Inflammation and Electron transport were enhanced in the mutant hearts,implying that these pathways might contribute to the cardiac remodeling resulted from Smad4 knock-out.
