Macrophage migration inhibitory factor antagonist,ISO-1,lead to cell cycle arrest in G2/M and facilitate migration in CHO cells
【摘要】:Macrophage migration inhibitory factor(MIF) plays divergent roles in different cell types and tissues.However,the function of MIF in Chinese hamster ovary cells(CHO) is still far from clear.In this study,the migration and cell cycle characteristicsof MIF were investigated in CHO cells using the specific MIF antagonist S,R-3-(4-hydroxyphenyl)-4,5-dihydro-5-isoxazole acetic acid methyl ester(ISO-1) by the scratch assay,immunohistochemistry and WB.MIF was expressed in CHO cells.ISO-1 could markedly enhance the cell migration by the scratch assay.The results of immunocytochemistry showed that the ISO-1 contributed to cell cycle arrest in G_2/M in MIF-knockdown cells.The results also demonstrated that ISO-1 might be involved in the AKT/GSK-3β pathway during the process of G_2/M cell cycle arrest.These results partially lay a theoretical foundation for better understanding the biological roles of MIF.
