Characteristics of immortal goat endometrial epidermal cells and endometrial stromal cells
【摘要】：正(Objective) To obtain available and convenient goat endometrial cells for in vitro studies,we established immortal goat endometrial epidermal cells(EECs) and endometrial stromal cells(ESCs).(Method) The human telomerase reverse transcriptase(hTERT) gene was introduced into primary goat EECs and ESCs,and got two immortalized cell lines, hTERT-EECs and hTERT-ESCs.After hTERT introduction,a series of biological characteristics of these cells were detected and compared with the primary cells.(Result) The results showed that the immortalized hTERT-EECs and hTERT-ESCs retained common morphological characteristics,such as normal cell cycle controls,anchorage-dependent contact inhibition and serum-dependent proliferation.And they had similar growth rates to the primary cells.In addition,these cells possessed normal karyotype,retained responsiveness to sex-steroid hormones and lacked transformed phenotypes on soft agar or in nude mice. (Conclusion) In summary,the immortalized goat endometrial cell lines(hTERT-EECs and hTERT-ESCs) had been developed what were karyotypically,morphologically and phenotypically similar to the primary parent cells.To date,hTERT-EECs had been passaged for 55 times and hTERT-ESCs for 60,and they would be convenient and consistent resources for further in vitro work.
