Differentiation of Ustilago esculenta may relate to phenotypic difference in Zizania latifolia

【摘要】:Ustilago esculenta,a pathogenic basidiomycete fungus,infects Zizania latifolia to form edible galls named jiaobai.Distinct growth conditions of U.esculenta resulted Z.latifolia to form three different phenotypes,named male Jiaobai,grey Jiaobai and white Jiaobai respectively.In this study,the sexual compatible haploid strains UeT14/UeT55 from grey Jiaobai(T-type strain) and UeMTlO/UeMT46 from white Jiaobai(MT-type strain) were respectively isolated and the life cycle without teliospores formation was carried out in vitro,which showed similarity to Ustilago maydis.Also,studies indicated that the T- and MT-type strains had great differences in many aspects.Firstly,spore germination rate in T-type strain is higher than the MT-type.Secondly,different carbon and nitrogen sources had significandy diverse influence on the growth of the fungus.Overall,T-type haploid strains showed faster growth rate and higher growth mass than MT- type haploid strains.But also there were some differences in the assimilation of carbon and nitrogen sources between the two type strains,that compaired to T-type strains,MT-type strains preferred NO_3~- in the inorganic nitrogen sources and had worse utilization of galactose.Thirdly,T- and MT-type strain had different behavior in haploid fusion and hyphae growth with external signals such as carbon or nitrogen sources,hydrogen peroxide(H_2O_2),pH and hydrophobicity.In general,MT-type strains were more sensitive to external signals compared with T-type,but there were exceptions that arginine and sodium glutamate had obviously inhibition effect on T-type but not MT-type,and also 0.5%(w/v) H_2O_2 promoted the filamentous formation in MT-type but inhibited that in T-type.Finally,genome sequencing analysis showed that there are multiple mutations in pathogenic genes of MT-type strains,in accordance with their reduced pathogenicity when compaired to T-type strains.Above all showed that there were obvious differentiations of U.esculenta from difference phenotypic Z.latifolia,indicating the relationship between U.esculenta strains differentiations and their host phenotypes.
