Accurate quantitation of astaxanthin from Haematococcus crude extract by spectrophotometery

【摘要】:正To establish a method for accurate quantitation of astaxanthin from Haematococcus by spectrophotometery based on the method which Boussiba et al.first reported,the influence of alkali treatment on astaxanthin and the optimal working wavelength for measuring of astaxanthin were investigated.We found that the carotenoids content declined about 25%in Haematococcus fresh cysts and up to 30%in the dry powder of cell wall broken cysts after alkali treatment.We also found that the dimethyl sulfoxide extracted chlorophyll of green Haematococcus cells barely absorbs at 520-550 nm.However, an unknown interference which made the results higher existed at 540-550 nm during the analysis of astaxanthin.Interestingly,we found that at 530 nm a good linear relationship existed between absorbance and astaxanthin content,and the influence of chlorophyll,other carotenoids,and an unknown interference were avoided.Therefore, with the 530 nm as a working wavelength,the alkali treatment is not necessary.The astaxanthin contents of two samples were measured at 530 nm.The results,which reflected the actual astaxanthin contents in the samples,were 6.93%and 11.96%lower than those at 492 nm,respectively.The data suggested that 530 nm is the most suitable wavelength for determination of astaxanthin in Haematococcus crude extract by spectrophotometery.
