A Meta-analysis About CircRNAAs the Biomarker of Gastric Cancer

【摘要】:Background:Gastric cancer was a malignant tumor in the world with the fifth incidence and third mortality and circRNA has close connect with it. however, it’s not clear how circRNA affects the gastric cancer. We performed a systematic review and meta-analysis to evaluate the relationship between circRNA and gastric cancer, aiming to identify circRNA can be used as a biomarker for the diagnosis of gastric cancer.Methods:We conducted a comprehensive search of PubMed, Web of Science and Embase for published experimental studies about circRNA as a diagnostic biomarker for gastric cancer from 2013 to June 2018. Two investigators independently reviewed the included studies and extracted relevant data. Sensitivity, specificity,area under curve (AUC), positive likelihood ratios, negative likelihood ratios, and diagnostic OR (DOR) were calculated to evaluate the diagnostic efficacy by STATA 12.0. The credibility is expressed in 95%confidence intervals (95%CI). Q Test and I2 are used for heterogeneity test, and subgroup analysis is performed at the same time.Results:Our final analysis included 11 studies,including 12 kinds of circRNAs, 11 in tissues, 5 in plasma, and all down regulated. Their pooled sensitivity, specificity, positive likelihood ratios, negative likelihood ratios, diagnostic OR and AUC with their 95%confidential intervals (95%CIs) were 0.713 [0.630, 0.784], 0.779 [0.701,0.842], 3.232 [2.463, 4.242], 0.368 [0.292, 0.463], 8.778 [6.108, 12.614] and 0.81 [0.78, 0.84]respectively. Subgroup analysis revealed that the sensitivity, specificity, positive likelihood ratios,diagnostic OR and AUC of circRNAs in plasm were higher than tissues but negative likelihood ratios. It’s also same in subgroup with different sample sizes (more than 100 and less than 100).The heterogeneity of the literatures was mainly due to the different backgrounds of gastric cancerand the differences in experimental design and operation process. And the Deeks’ funnel map revealed there was no obvious public bias in the literature.Conclusion:CircRNAs have high sensitivity and specificity in the diagnosis of gastric cancer, and it may become an auxiliary diagnostic biomarker of gastric cancer.
