Mechanism of Microbeam Irradiation Induced Bystander Effect

【摘要】:正Purpose: Radiation induced bystander effects may play an important role in cancer risks associated with environmental, occupational and medical exposures and they may also present a therapeutic opportunity to modulate the efficacy of radiotherapy. However the mechanisms underpinning these responses in tumor and normal cells are poorly understood. This study investigated the signalling factors involved in the bystander response of chromosome damage in the cell lines with different genotypes. Methods: The development of microbeams has allowed individual cells to be targeted at specific subcellular locations with precise doses of radiation. The charged particle microbeam facility of Gray Cancer Institute was used to deliver a desired number of helium (3He2+) particles to either nuclei or cytoplasm of a part of cells within a population of either human glioblastoma T98G or primary human fibroblasts AG01522, then the chromosome damage of micronuclei (MN) formation in nonirradiated bystander cells was measured, and the relative signalling factors of nitric oxide (NO), reactive oxygen
