Aggravated Pulmonary Injury after Subarachnoid Hemorrhage in PDGF-B~(ret/ret) Mice

【摘要】:Background: Recent advances in surgical and neuroprotective strategies could effectively manage the pathophysiological progresses of SAH. However, pulmonary dysfunction frequently occurred in SAH patients with an increased risk of unsatisfied outcomes. Methods: In the present study, PDGF-B retention motif knockout mice(PDGF-B~(ret/ret)) and adeno-associated virus PDGF-B were employed. Neurological score, SAH grade, HE staining, PaO_2/FiO_2 ratio, Western blotting and Immunofluorescence staining were performed to evaluate the neurological deficits and pulmonary functions in endovascular perforation SAH models. Results: We found neonatal PDGF-B~(ret/ret) mice exhibited pulmonary atelectasis at 12-hours after birth. Further investigation showed decreasing in PaO_2/FiO_2 and lung-specific surfactant proteins in adult PDGF-B~(ret/ret) mice. These dysfunctions were much worse than wildtypes at 24-hours after SAH. And PDGF-B overexpressing could alleviate the pulmonary dysfunctions after SAH. Conclusions: These results suggested pulmonary dysfunction after SAH, as well as involvement of PDGF-B signal for the pathophysiological process and future therapeutic targets of the pulmonary injury treatment after SAH. Further studies are still needed for the pathophysiology investigations and translational researches on the pulmonary injuries after SAH.
