EGF activation of p21 activated kinase-1(Pak1) is critical for human amnion mesenchymal stem cell migration in vitro
【摘要】:Whether epidermal growth factor(EGF) induces growth and directional migration of human amnion mesenchymal stem cell(hAMSC) remains unclear.In the current study we examined the effect of EGF on proliferation and directional migration of hAMSC in vitro.The movement of EGF-stimulated hAMSCs was monitored over a two-hour period using Dunn chemotaxis chamber.A linear concentration gradient of EGF-stimulated hAMSCs showed special directional preferences with migration at a mean rate of 6.45 μm/h.In addition,we demonstrated that EGF increased epidermal growth factor receptor(EGFR),protein kinase B(AKT),and extracellular signal-regulated kinase-1/2(ERK) phosphorylation levels.Especially,EGF-induced activation of ERK was blocked by AG1478(EGFR inhibitor) and LY-294002(phosphoinositol-3 kinase(PI3 K) inhibitor).We also investigate whether EGF regulates Pak1 signaling and is critically linked to hAMSC migration.Moreover,EGF promoted directional migration of hAMSCs,which was attenuated by AG1478,LY-294002,and U0126(ERK inhibitor).In conclusion,our data demonstrated that EGF-induced proliferation and migration of hAMSCs were mediated by EGFR,which activated PI3 K/AKT and ERK signals in hAMSCs.
