Interaction of pro-inflammatory cytokines and angiotensinⅡin paraventricular nucleus in regulating sympathetic activity and cardiac sympathetic afferent reflex in rats

【摘要】:正Cardiac sympathetic afferent reflex(CSAR) is a sympatho-excitatory reflex and can be induced by the stimulation of the cardiac sympathetic afferents with bradykinin(BK),adenosine,and hydrogen peroxide released from the myocardium during myocardial ischemia.The enhanced CSAR partially contributes to the sympathetic activation in experimental hypertension and chronic heart failure.The present study was designed to determine the interaction of pro-inflammatory cytokines and angiotensin II in paraventricular nucleus(PVN) in regulating the sympathetic activity and CSAR.The rats were anesthetized with urethane andα-chloralose and underwent bilateral sinoaortic denervation and cervical vagotomy.The levels of tumor necrosis factor-alpha(TNF-α) and interleukin- 1 beta(IL-1β),two kinds of pro-inflammatory cytokines,were determined with ELISA.The CSAR was evaluated by the response of renal sympathetic nerve activity(RSNA) to epicardial application of BK.Epicardial application of BK increased the levels of TNF-αand IL-1βin the PVN,which was abolished by the pretreatment with the PVN microinjection of a cytokine blocker pentoxifylline(PTX).Microinjection of TNF-αor IL-1βinto the PVN increased the RSNA and mean arterial pressure(MAP),and enhanced the CSAR,which were attenuated by pretreatment with PTX or an AT_1 receptor antagonist losartan.The PVN microinjection of PTX abolished the CSAR but had no significant effect on the baseline RSNA and MAP,while losartan not only abolished the CSAR but also decreased the baseline RSNA and MAP.The low dose of TNF-αor IL-1βcombined with angiotensin II in the PVN caused much greater responses than the same dose of TNF-α,IL-1βor angiotensin II alone.These results indicate that stimulation of cardiac sympathetic afferents increases TNF-αand IL-1βlevels in the PVN.TNF-αand IL-1βin the PVN enhance the CSAR and sympathetic activity,which is involved the interaction between these pro-inflammatory cytokines and angiotensin II in the PVN.
