Detection of bacterial DnaK and enolase specific antibodies in the serum of patients with chronic rhinosinusitis with nasal polyps(CRSwNP)

【摘要】:Background: The statuses of the antibodies against heat shock protein 70(HSP70) and enolase in chronic rhinosinusitis(CRS) are unclear yet. We hypothesized that concentrations of antibodies against bacterial DnaK and enolase increased in the serum of patients with CRS.Objective: To detect bacterial DnaK-and enolase-specific antibodies in the serum of patients with chronic rhinosinusitis with nasal polyps(CRSwNP) Methods: Molecular biology and ELISA techniques were employed to obtain recombinant proteins encoding the DnaK and enolase of Streptococcus pneumonia(S.P) and to measure the existence of IgG and IgE antibodies against these molecules in sera of participating subjects.Results: The concentrations of specific IgG antibodies against the DnaK and enolase of S.P were elevated in the sera of patients with CRS or allergic rhinitis(AR) although only the levels of DnaK-specific IgG were significantly higher than those of the controls(p 0.05). The concentrations of enolase-specific IgE were significantly greater in the serum of AR than that of the control. The levels of IgG and IgE antibodies against DnaK in the CRSwNP group and enolase in the AR group were positively correlated with each other. Up-to 60% of the antigen-antibody specificity was neutralized by using whole proteins extracts derived from Staphylococcus aureus, suggesting the high homology of these proteins between different bacterial species. Conclusions: Our data suggest that the presences of bacterial DnaK-and enolase-specific IgG and IgE antibodies in the serum of CRS and AR and the high homology of these proteins between different species raise the possibility that these antigens and antibodies may play important roles in the pathogenesis of the disease through "molecular mimicry".
