The Hallmarks of Cancer: LncRNA Point of View

【摘要】:Precise regulation of signaling is crucial for cell growth,differentiation,metabolism,apoptosis,and organ formation.Dysregulation of cell signaling can lead to the initiation and progression of cancer.For example,the PI3 K-AKT pathway is hyperactivated in many human cancers;several drugs that inhibit this pathway are currently being tested in various preclinical and clinical trials.We,as well as others,have shown that pharmacologic inhibition of the PI3 K-AKT pathway results in feedback activation of other oncogenic signaling pathways.A point-of-view that includes IncRNAs in the study of breast cancer biology could be the conceptual advance that allows for such breakthroughs.Metabolism reprogramming is a hallmark of cancer that allows for uncontrolled proliferation and survival during tumor progression.The Warburg effect is established on the observation that tumor cells implement abnormally high rates of glucose uptake to meet their increased energy,biosynthesis,and redox needs.This finding serves as the basis for a new strategy for cancer treatment that targets cancer metabolism.We hypothesize that by focusing on novel lncRNA targets to block the PI3 K/AKT signaling pathway,cancer cell metabolism and tumor progression can be inhibited.Also,we will be able to expand the arsenal of targeted therapies and therefore individualize cancer treatment.In the succeeding study,we will conduct an in-depth investigation of the Lipid-LncRNA-Kinase pathway,which is important for tumor growth and survival.In this proposed study,we will focus on our recent discoveries of novel oncogenic function of the lipid interacting IncRNAs in cancer metabolism and the role of glucose-mediated energy homeostasis in the regulation of AKT or other potential metabolic pathways,such as HIF or Myc signaling.This will be detected by innovative RNA pull-down methods coupled with mass spectrometry.Our objective is to assess the mechanistic cross-talk between glucose metabolism and the Lipid-LncRNA-Kinase pathway with the goal of identifying new biomarkers or ncRNA drug targets in this pathway for breast cancer treatment.Ultimately,the success of our study will lead to a better understanding of the important Lipid-LncRNA oncogenic pathway.The identification of LncRNA-Lipid interactions,a new mechanism of IncRNA action,is of great interest and would broaden the spectrum of IncRNA functions to include their novel role as essential mediators of signaling pathways,which would expand our understanding of the role of IncRNAs in homeostasis and disease.
