HMGB3 modulates ROS production via activating TLR cascade in Apostichopus japonicus

【摘要】:High mobility group box protein 3(HMGB 3) was found to play a significant role in regulating proliferation and inflammatory response in vertebrates.However,the functional roles of HMGB3 in invertebrate were largely unknown.In current study,the HMGB3 homologue molecule was identified from A.japonicus(designated as AjHMGB3) by RACE approach.The full-length cDNA of AjHMGB3 was of 2298 bp with an open reading frame of 1320 bp encoding a 439 amino acid residue protein.Structural analysis revealed that AjHMGB3 processed the two conserved characteristic HMG boxes(133-204 aa and 210-279 aa) and an acidic tails.Multiple sequence alignment and phylogenetic analysis together supported that AjHMGB3 belonged to a new member of HMGB3 protein subfamily.Spatial expression analysis revealed that AjHMGB3 was expressed in all examined tissues other than in tentacle with the larger magnitude in intestine.The Vibrio splendidus challenge in vivo and LPS stimulation in vitro could both significantly up-regulate the mRNA expression of AjHMGB3 in coelomocytes,which were consistent with the expression profiles of TLR cascade members.We further investigated the expression profiles of AjMyD88,Ajp105 in AjHMGB3 over-expressed coelomocytes and suppress-expressed,and found that AjMyD88 and Ajp105 were up-regulated to2.19-fod and 2.83-fold in over-expressed group,and down-regulated to 0.38-fold and0.43-fold in suppress-expressed group compared to the control group,respectively.Consistently,the p50 subunit was also showed induced expression by western blot analysis in over-expressed group and down-regulated expression in suppress-expressed group.In the same condition,the respiratory burst was evaluated by 37.5%in over-expressed group and eliminated by 28.2%in suppress-expressed group.Our present findings collectively suggested that AjHMGB3 acted as NF-κB activator and produced ROS production in sea cucumber.
