AQP2 is regulated by estradiol in human endometrium and involved in spheroids attachment in vitro
【摘要】：Background:17β-estradiol(E2) and aquaporin 2(AQP2) are associated with endometrial receptivity.E2 can directly regulate AQP2 expression in endometrial cancer cells.This study aimed to assess the role of AQP2 in embryo implantation.Methods:Normal endometrial samples were collected from women who attended the Zhejiang Women's Hospital for in vitro fertilization and embryo transfer(IVF-ET) treatment.Women with endometrial abnormalities were excluded.Samples were categorized as early-mid proliferative,late proliferative,early secretary,mid-secretary,and late secretary phases.AQP2 mRNA(qRT-PCR) and protein(western blot) expression in response to E2 was assessed in normal human endometrium.The effects of AQP2 on spheroid attachment were assessed by an in vitro co-culture assay siRNA against AQP2 Results:The highest levels of AQP2 were observed during the late proliferative and mid-secretory phases.The lowest levels of AQP2 were observed during the early proliferative and late secretory phases.Both 10~(-9) and 10~(-7) M E2 for 24 h up-regulated AQP2 in cultured endometrium.Knock-down of AQP2 by siRNA significantly decreased JAr spheroids attachment,with or without E2(P0.01).Conclusion:AQP2 expression in human endometrium is mediated by estrogen.Low AQP2 level could be a potential cause for impaired uterine receptivity.
