Comparative study of hypnotic effects of jujubosides and flavonoids isolated from Semen Ziziphi Spinosae
【摘要】：正Semen Ziziphi Spinosae,the seed of Z.jujuba Mill.(Rhamnaceae),has been widely used in treating insomnia and anxiety.The present study further investigated the hypnotic effects of two main components of semen Ziziphi Spinosae,flavonoids and jujuboside,under both dark and light time.The results of this study suggested several facts as followed.Jujuboside significantly dose-dependently augmented pentobarbital(45 mg/kg,i.p.)-induced sleep,proved by increased sleep time and such an effect was potentiated by 5-hydroxytryptophan(5-HTP,2 mg/kg,i.p.).Pretreatment with p-chlorophenylalanine(PCPA,300 mg/kg,i.p.),an inhibitor of tryptophan hydroxylase,significantly decreased pentobarbital-induced sleep time,and jujuboside significantly reversed this effect.Both jujuboside and flavonoids can significantly increase the sleep time on normal rats during dark and light time.However the influences on the sleep parameters are not same.Flavonoids evidently increased all the sleep parameters during both light time and dark time.During light time,jujuboside considerably increased the total sleep time and REM sleep time while had no significant effects on the rest sleep parameters.During dark time jujuboside significantly increase the total sleep time,NREM sleep time and the light sleep while had no significant effects on deep sleep time and REM sleep time.The results suggested that the 5-HT receptor system may involve in the hypnotic effect of jujuboside on pentobarbital treated mice and the hypnotic effect of jujuboside on normal rats may be influenced by circadian rhythm.
