Identification,classification and expression analysis of KNOX gene family in whole genome in Pyrus bretschneideri cv.Dangshan Su

【摘要】:Pyrus bretschneideri cv.Dangshan Su is a Chinese pear variety widely grown across China.However,the fruits of Dangshan su pear with higher stone cell content compared with other pear varieties,which resulting in poor taste,impaired nutrients and severely affected its nutritional value and economic benefits.Therefore,the reducing of stone cell content is essential for improving the quality of Dangshan pear fruit.The formation of stone cells has been shown to be closely related to the synthesis,transport and deposition of lignin.Knottedl-like homeobox(KNOX) proteins are homeodomain transcription factors that play an important role in plant lignin metabolism and meristem development.However,at present,there is no report on the study of pear KNOX family,and it is not clear which members are involved in lignin biosynthesis.In this study,18 KNOX family members were identified from the genome of Dangshan pear and PbBP was found to be orthologous with AtBP by phylogenetic analysis,multiple sequence alignments and temporal and spatial expression patterns analysis.Fluorescent quantitative analysis showed that this gene might negatively regulate lignin metabolism in pear.Then on the base of these,this study cloned the promoter sequence of COMT and CCOMT{key enzyme genes for lignin synthesis in pear) and constructed yeast one hybrid vector.It was found that PbBP could be combined with some promoter sequence of COMT" and CCOMT.And then we will use the corresponding Arabidopsis mutants for functional complementation studies to further investigate the role of PbBP in lignin biosynthesis.In this study,we studied the molecular evolution of pear KNOX family and identified members of KNOX gene family related to lignin metabolism of pear fruit.At the same time,which not only provide new insights into the role of KNOX family in lignin metabolism of pear,but also lays a foundation for molecular regulation of lignin synthesis and fruit quality improvement in pears.
