Genome-wide profiling of lysine butyrylation reveals positive regulation of transcription in rice

【摘要】:Histone post-translational modification(PTM) plays a vital role in altering chromatin structure and influences gene expression in eukaryotes.Recent years,more and more novel PTMs have been identified.Lysine butyrylation(Kbu) is a new histone modification that catalyzed by acetyltransferases and dynamically competing with acetylation in controlling gene expression in mammalian.However,lysine butyrylation haven't been reported in plants.Besides,few researches took a look at it with high-throughput methods.Here we firstly genome-wide profiled lysine butyrylation with high resolution in rice(Oryza sativa).Chromatin immunoprecipitation sequencing(ChlP-seq) results showed a major enrichment of Kbu in genebody region(71.39%),especially in exon region.An obvious positive correlation between Kbu and gene expression levels were also observed in combination with RNA-seq analysis.Next,we integrated DNase-seq and other 12 histone modifications public data in rice,to explore the concurrence among DNase-Hypersensitive sites(DHS) and all these histone modifications.Similar profiles were detected between histone Kbu and several active histone modifications with overlapping ratio from 74%to 96%,suggested that Lysine butyrylation is an active mark of transcription in rice.Furthermore,GO enrichment analysis indicated that histone Kbu is involved in diverse cellular processes,especially in cytoplasm and membrane.Collectively,all these results revealed that lysine butyrylation can positive regulate gene expression and our researches will provide plenty of resources for further studies on histone Kbu and other epigenetic regulations in plants.
