A Receptor-like Protein Gene Gbvdr6 of Cotton(Gossypium barbadense),confers resistance to Verticillium wilt in upland cotton and is Involved in the Jasmonic Acid and Ethylene Signaling Pathway

【摘要】:Verticillium wilt is a soil-borne disease that can cause devastating losses in cotton production.The effective way to control Verticillium wilt is through genetic improvement because there is no effective chemical means to combat the disease.Gossypium barbadense is usually resistant or tolerant to Verticillium wilt.The genetic improvement of G.hirsutum through introgression from G.barbadense is an effective way to control Verticillium wilt.However,hybrid breakdown and sterility in interspecific crosses between G.hirsutum and G.barbadense are real obstacles to the efficient use of Verticillium wilt resistance from G.barbadense.Therefore,the identification of disease-resistance genes of G.barbadense will benefit efforts towards the genetic improvement of cotton resistance to Verticillium wilt.Here,a receptor-like protein coding gene,Gbvdr6,that was isolated from Verticillium wiltresistant cotton G.barbadense cultivar Hai7124.Sequence mapping through a Blastn search in the upland cotton(G.hirsutum) cv.TM-1 genome revealed that Gbvdr6 had two homologs in the G.hirsutum.One was in the AOl(Cl) chromosome,and the other was in the DOl(cl5) chromosome.The gene was located in the gene cluster containing another 2 receptor like proteins Gbvel,Gbvdr5 and adjacent to the Verticillium wilt-resistance QTL hotspot.Gbvdr6 was induced by Verticillium dahliae Kleb and plant hormone Salicylic acid(SA),methyl jasmonate(MeJA) or ethephon(ETH).Subcellular localization analyses indicated that Gbvdr6 was localized to the plasma membrane.The over-expression of Gbvdr6 in Arabidopsis and cotton enhanced resistance to V.dahliae.Moreover,the over-expression of GbvdrS significantly elevated the expressions of JA/ET signaling pathway-related genes such as PR3 and PDF 1.2 and the transgenic plants were less sensitive to MeJA.Furthermore,the accumulation of reactive oxygen species and callose was triggered at the early time points of V.dahliae infection.These results suggest that Gbvdr6 conferred resistance to V.dahliae and accompanied with increased expression of the JA/ET signaling pathway-related genes.
