Ca~(2+) signaling in embryonic stem cells fate decision and in derived cardiac lineages

【摘要】:Embryonic stem cells(ESCs), derived from the inner cell mass of preimplantation embryo, are pluripotent cells having the ability to differentiate into derivatives of all three germ layers(endoderm, mesoderm, and ectoderm) in vitro. ESCs recapitulating the early stages of lineage differentiation represent an appropriate model to study the regulatory mechanisms underlying early embryogenesis besides their usage in tissue repair. Intracellular Ca~(2+) signals have crucial roles in the regulation of various cellular processes, including proliferation, differentiation, and apoptosis, and myocyte contraction. Inositol 1,4,5-trisphosphate receptors(IP_3Rs) and ryanodine receptors(Ry Rs) are two major intercellular Ca~(2+) release channels located on the internal Ca~(2+) store endoplasmic reticulum and have important roles in the regulation of cellular behaviors through generation of spatially and temporally signal patterns of cytosolic free Ca~(2+) concentration. However, the precise function of Ca~(2+) signals and the regulation of IP_3Rs/Ry Rs in specific lineage fate decision of ESCs and ESC-derived early cardiomyocytes remain largely unknown. To understand them, we studied the roles of the above channel-mediated Ca~(2+) signals during the cell fate decision of ESCs towards mesodermal cells, cardiac progenitors, cardiomyocytes as well as neuronal cells. Our data suggest that Ca~(2+) signals may play more important roles in the cell fate decisions during early ESC differentiation and in the maintaining physiological function of early ESC-derived cardiomyocytes than expected.
