Genetic variability and molecular evolution of the untranslated regions of Potato virus Y

【摘要】:Isolates of Potato virus Y(PVY) differ widely in their biological properties.These properties may depend on the structure of viral RNA populations comprising the different isolates.As a first approach to study the molecular basis of the biological variability,we have compared the sequences of multiple cDNA clones of the two terminal regions of the RNA from different PVY isolates.In this study,the Genetic variability and molecular evolution of PVY in their untranslated region were studied using 270 sequences generated from isolates collected from potato in China and 148 PVY sequences selected from "GeneBank" which classification,country and continent origins have been mentioned.High genetic differentiation was found in the populations from the different countriesgrouped by continent,with higher nucleotide diversity in Europe thanAmerica,Asia and the isolates collected in china.We demonstrated that although there is high variability at the 5'-UTR among members of the 4 major groups of PVY isolates,this region is mainly conserved among individual members within each group andat the 3'-UTR were more diverse and variable over the host species and the regions where they were isolated.Further analyses showed that spatial genetic structure in the untranslated region of PVY populations was likely caused by demographic dynamics of the pathogen and natural selection generated by habitat heterogeneity.Isolates from all groups tended to group together,indicating that nucleotides diversification was maintained,in part,by host-driven adaptation.
