A Phytophthora RxLR Effector Manipulates Host Immunity by Regulating SAGA-mediated Histone Acetylation Modification

【摘要】:Plant pathogens secrete a large number of effector proteins to subvert host cell immunity.However,knowledge of the mechanism on how effector proteins manipulate host immunity remain largely unknown.Epigenetic modifications,such as histone modification,serve as important strategy of host innate immunity against various pathogens.Nevertheless,pathogens secretes effector proteins to suppress host histone modification pathways.Here,we shows that PsAvh23,a virulence-essential effector from the soybean stem and root rot pathogen Phytophthora sojae,interacts with plant transcriptional coactivator protein ADA2b(alteration/deficiency in activation),a subunit of SAGA complex.The SAGA complex recruits the catalytic subunit Gcn5(general control nonderepressible 5) via ADA2 b to regulate histone acetylation modification.We found that PsAvh23 competes with soybean Gcn5a(Gcn5 subunit of G.max) and depletes it out of ADA2b-Gcn5 subcomplex of SAGA complex,resulting in a significant reduction of histone acetylation and H3K9 ac in vitro and in vivo.In addition,we demonstrated that the PsAvh23 down-regulated a subsets of defense-related genesand thisrequires SAGA-mediated histone acetylation.Our findings reveal a novel mechanism that plant pathogens can employ effector proteins to regulate host epigenetic modifications,such as histone acetylation modification,as a general virulence strategy to counteract host defenseat an epigenetic level.
