Study on Genetic Diversity of Fusarium pseudograminearum Populations Causing Wheat Crown Rot by ISSR Analysis

【摘要】:Crown rot of wheat caused by Fusarium pseudograminearum,which was first detected in Queensland in 1951.Li et al reported the F.pseudograminearum causing crown rot for the first time in China in 2012.It is serious increasingly in winter wheat area of Huang-Huai region of China and affects yield-increasing of wheat in the recent years.The genetic diversity of F.pseudograminearum in HuangHuai region of China have not been reported.The aim of the present study was to examine the population structure of eight populations of F.pseudograminearum collected from different regions in Huang-Huai by using ISSR-PCR,and to characterize the genetic differentiation within and between these populations.An understanding of the genetic structure of F.pseudograminearum may provide theoretic foundation and technique measures for the management of wheat crown rot Seventeen ISSR primers which can amplify more polymorphic loci were screened from 97 ISSR primers.The genetic diversity of 172 F.pseudograminearum isolates from Henan,Hebei,Shandong and Shanxi province was analyzed with this 17 primers.The amplification results showed that 234 fragments were amplified;polymorphic loci were 218 which accounted for 93.16%in the total amplified fragments.The average number of bands per primer was 13.76 and the size of the amplified fragments ranged from 150 bp to 2500 bp.The POPGENE analysis results showed that the coefficient of population genetic differentiation(Gst) among geographical groups was 0.2229,while it reach 0.7771 within the populations.The genetic diversity within the populations was much more abundant than among the groups.The number of migrants per generation between eight geographical populations was 1.7434,which indicated that genetic information exchange was frequent between the eight groups.The dendrogram based on ISSR markers revealed that eight geographical populations were clustered into 3 clusters at the threshold of genetic similar coefficient 0.95.The grouping results indicated that the genetic variation of F.pseudograminearum was related with its geographic distribution.Thus,this techique can be used to analyze the genetic diversity of F.pseudograminearum in the future study.
