microRNA-133b regulates zebrafish axon regeneration of Mauthner-cell at a single cell level through targeting tppp3
【摘要】:Axon regeneration, which relies on rapid changes in gene expression, is fundamental to nerve repairing and functional recovery, indicating the involvement of micro RNAs(miRNAs). Mir-133 b have been proved to play an important role in regeneration in zebrafish, but its role in regulating axon regeneration in vivo is still controversial. Here, combing single-cell electroporation essay with vector-based expression system, we regulated the expression of miR-133 b in Maunthner-cells(M-cells) at the single-cell level. Our results find out that overexpression of miR133 b can inhibits axon regeneration, whereas inhibition of miR-133 b can promote axon regeneration. Moreover, we found out that the function of miR-133 b during axon regeneration by directly targeting Tppp3, a member of the Tubulin polymerization-promoting protein family that induces tubulin polymerization. Gain or loss-of-function of tppp3 experiments indicate that tppp3 is a novel gene that regulates axon regeneration. What's more, we observed a reduction of mitochondrial motility in miR-133 b overexpressed M-cells. Taken together, we extend the role of miR-133 b in axon regeneration in zebrafish at single-cell level and find a new regeneration-associated gene tppp3.
