Inflammation imaging and investigation of drug-induced hepatotoxicity with reaction based fluorescent probes for ONOO-
【摘要】：Peroxynitrite (ONOO~-) is a kind of reactive oxygen species (ROS) with super activity of oxidization and nitration, and overproduction of ONOO-is associated with pathogenesis of many diseases. Thus, accurate detection of ONOO~- with high sensitivity and selectivity is imperative for elucidating its functions in health or disease states. Herein, we present some reaction based fluorescent probes (e.g.Mito-CC, Rhod-ONOO~-) for ONOO~- by combining rational design strategy and dye-screening approach[1-3]. These probes showed highly sensitive and selective response to ONOO-under physiological conditions, and were successfully used for imaging of ONOO~- in an inflamed mouse model and elucidating the drug-induced hepatotoxicity pathway, as well as real-time monitoring of hepatotoxicity in living animals. Therefore, these probes can be potential biological tools to explore the roles of ONOO~- under different physiological and pathological settings.
