The Optimization Studies on Determination of Astragaloside Ⅳ in Astragali Radix

【摘要】：Objective To optimize the test solution processing method for determination of astragaioside IV to establish a UPLC method for determining the contents of astragaioside IV in astragali Radix.Methods To Use the optimized method:ultrasonic extraction,N-butanol isolation after methanol and ammonia mixed;determination of the astragaioside IV by UPLC-ELSD.Chromatographic conditions:Agilent poroshell 120 EC-C_(18) Column(50 mmx4.6 mm,2.7 urn),Acetonitrile-water(32:68) as the mobile phase,Column temperature 30 °C,flow 0.5 ml·min~(-1),ELSD detector.Results The astragaioside IV content of the same 11 batches of samples by the optimized method in this study is significantly higher than one by the "Chinese Pharmacopoeia" 2010 edition method.Conclusion:The optimized method in the study is fast,complete saponification,smaller loss and good repeatability,which can be suggested to improve the method in Chinese Pharmacopoeia(2010 edition).
