Insights into differentiation of stem into the adult Leydig cell lineage

【摘要】:Adult Leydig cells(ALCs) are the differentiated steroidogenic cells in the testes that produce testosterone.ALCs develop postnatally from a pool of stem cells,referred to as stem Leydig cells (SLCs).SLC is a spindle-shaped cell that lacks Leydig cell markers,including luteinizing hormone receptor and 3|3-hydroxysteroid dehydrogenase(HSD3B).The commitment of SLC to the progenitor Leydig cell(PLC),the first of the cell that has been characterized to enter the Leydig cell lineage, requires growth factors,including Dessert Hedgehog and platelet-derived growth factor-AA.PLC is steroidogenic and produces mainly androsterone.The next transition is from PLC to the immature Leydig cell(ILC).This transition requires luteinizing hormone and androgen.ILC is an ovoid cell that is competent for androgen production,producing primarily androstanediol.The final transition in the Leydig cell lineage is the differentiation into ALC.This step involves the reduced expression of the androgen metabolizing enzyme 5α-reductase 1.ALC produces testosterone as its final product. The transitions in the Leydig cell lineage are associated with the progressive down-regulation of the proliferative capacity of the involved cells,and the up-regulation of testosterone biosynthesis and many factors regulate these transitions.
