NLRP4F,a novel component of the subcortical maternal complex,is involved in mouse maternal to zygotic transition
【摘要】:Recently,we have identified a subcortical maternal complex(SCMC) critical for mammalian maternal to zygotic transition.Although several proteins of the SCMC have been reported,other components are still needed to be investigated to fully understand the mechanism of this complex.Here,through analyzing the results obtained by co-immunoprecipitation and mass spectrometry,a novel component of the SCMC,NLRP4 F,was identified.Using ovarian lysates and 293 T expression system,we confirmed NLRP4 F could directly interact with MATER,TLE6,FLOPED and ZBED3,except Filia.NLRP4 F exhibited a maternal expression pattern and the absence of NLRP4 F leaded to decreased female fecundity and delay of embryogenesis.NLRP4 F null zygotes were asymmetric division because of the abnormal cytoplasmic F-actin reshwork.The distribution of organelles was disturbed in Nlrp4 f null zygotes and oocytes,resulting from the aberrant microtubules and cytoplasmic lattices,oocyte-and early embryo-specific structure.These phenotypes were also observed in the knockout mice of other known SCMC genes.In addition,NLRP4 F was unstable when any member of the SCMC was disrupted,while the absence of NLRP4 F did not affect the expression of other SCMC proteins.In conclusion,our study shows that NLRP4 F is a novel component of the SCMC and plays important roles during mouse maternal to zygotic transition.
