The Projection between Ventrolateral Preoptic Area and Tuberomammillary Nucleus in Sleep-Wakefulness Regulation of Rats

【摘要】:To investigate the effects of relationship between ventrolateral preoptic nucleus(VLPO)and tuberomammillary nucleus(TMN)on the regulation of sleep-wake cycle in rats.SD male rats were randomly divided into seven groups:night control group(TMN+ACSFVLPO+ACSF)and night experimental groups[(1)TMN+ACSFVLPO+L-Glu;(2)TMN+BicVLPO+ACSF;(3)TMN+BicVLPO+L-Glu];daytime control group(VLPO+ACSFTMN+ACSF)and daytime experimental groups[(1)VLPO+ACSFTMN+L-Glu;(2)VLPO+TripTMN+L-Glu].Compared with the night control group,the amount of wakefulness was decline along with the enhancement of NREM sleep and TST after L-Glu microinjected into VLPO,while there was no significant change in REM sleep.Microinjection of Bic into TMN in TMN+BicVLPO+ACSF group enhances rat wakefulness,however REM and NREM sleep had no significant changes.Compared to those of L-Glu microinjected into VLPO alone in the TMN+ACSFVLPO+L-Glu group,the wakefulness was enhanced,meanwhile,REM sleep,NREM sleep and TST were decreased after L-Glu microinjected into VLPO followed by the Bic microinjected into TMN.Compared with the daytime control group,the amount of wakefulness was enhanced along with the reduction of NREM sleep and TST after L-Glu microinjected into TMN,meanwhile there is no significant change in REM sleep.Compared to those of L-Glu microinjected into TMN alone in the VLPO+ACSFTMN+L-Glu group,the wakefulness was decreased,meanwhile,REM sleep,NREM sleep and TST were enhanced after L-Glu microinjected into TMN followed by the Trip microinjected into VLPO.We concluded that VLPO activation could inhibit the TMN neuronal activity via GABAA receptor and induce a sleep-promoting effect,moreover,TMN activation could inhibit VLPO neuronal activity via H1receptor and induce a wake-promoting effect.
