De Novo formation of arteries revealed by genetic lineage tracing

【摘要】:Arteriogenesis and de novo artery formation are two important concepts to explain collateral vessel development.Arteriogenesis means that collateral arteries are formed from preexisting arteries by in situ proliferation,while de novo artery formation indicates new arteries form from capillaries through remodeling and recruitment of smooth muscle cells.To date,these two models could not be distinguished clearly.We aim to distinguish these two models by Apln-Cre ER mediated genetic lineage tracing.APLN is specifically expressed in preliminary coronary endothelial cells(ECs).However,APLN is no longer expressed in ECs of coronary arteries(CAs)once they mature.By ligation of left descending CA of neonatal hearts,we established a model for robust new CAs formation.Through lineage tracing of capillary ECs but not arteries ECs,we showed that major collateral vessels around ligation site develop from preexisting non-labeled coronary arteries,indicating arteriogenesis.The medium-size CAs arise de novo by remodeling of pre-labeled capillaries,indicating de novo formation.MI induced more medium-size CAs generation by de novo formation model.Altogether,lineage tracing by Apln-Cre ERdistinguished two different models in CA formation after MI,experimentally demonstrating that arteriogenesis and de novo artery formation account for different compartments of collateral arteries.
