Construction of a public database for cancer-causing mutations in the Chinese population of HBV virus
【摘要】:Hepatitis B virus(HBV) infection is a global health problem, with approximately 250 million people worldwide suffering from chronic HBV infection.There are about 93 million infected people in China, of which about 30 million are chronic hepatitis B patients, and some patients may develop liver cirrhosis or even liver cancer.It has proved that the genetic characteristics of HBV strain subtypes in Chinese population and HBV susceptibility in Chinese population are different from those in Europe and America. However, most of the current studies are aimed at European and American populations or mixed populations, lack of systematic HBV strains susceptible to the Chinese population and Chinese population-specific HBV mutations, especially the organization and induction of oncogenic mutations.This study started with the candidate mutation sequences of HBV virus and hepatocarcinoma obtained from published literature and public databases, using open source bioinformatics tools such as netMHCPAN and PickPocket to predict the antigenic peptides produced by candidate HBV mutations and determine the MHC classin molecular sequence. And using molecular docking and molecular dynamics simulation to predict the binding affinity and stability of antigenic peptides of HBV virus mutations to host MHC steroids, and to analyze the immune escape mechanism of candidate HBV mutations in liver cancer.In addition, statistical methods were used to analyze the number, distribution and other information of HBV virus genome mutations and the corresponding sequence characteristics of liver cancer genome to summarize the corresponding relationship between HBV mutation and the occurrence and development of liver cancer. In the end, we will summarize the research results and construct a public database of key mutations in the carcinogenicity of HBV virus in Chinese population. This study will provide new clues for the selection of prevention, diagnosis and therapeutic targets for liver cancer.
