The genome sequence of Chinese Herb Erigeron breviscapus and complete biosynthesis of scutellarin in yeast
【摘要】:Erigeron breviscapus is a well-known traditional Chinese medicinal herb to treat cardiovascular and cerebrovascular diseases. Scutellarin is the main active ingredient of E. breviscapus. Farming supply of E. breviscapus resource is not enough to satisfy the requirement of increasing markets in China. A whole-genome sequencing of E. breviscapus, leading to a preliminary draft of the genome, was combined with bioinformatics analysis, enzyme discovery, pathway construction and strain optimization to realize full scutellarin biosynthesis in yeast starting from glucose. Two key enzymes Flavonoids 6-hydroxylase(F6H) and Flavonoid 7-O-glucuronosyltransferase(F7GAT) in the pathway were identified from E. breviscapus for the first time. After metabolic engineering and fed-batch fermentation, the final yields of scutellarin and another active ingredient apigenin-7-O-glucuronide respectly reached to 108 and 185 mg/L after the fed-batch fermentation. Our research realizes an alternative supply for these medically relevant compounds and will bring this traditional Chinese medicine benefit to the cerebrovascular patients around the world.
