Aptamer/polydopamine nanospheres complex based fluorescent biosensor for sensitive and selective detection of ochratoxin A

【摘要】:In this work,based on the high fluorescence quenching efficiency of polydopamine nanospheres(PDANS) and the specific recognition characteristic of aptamer,we construct a rapid,simple,sensitive and economical fluorescent biosensor for "turn-on" detection of ochratoxin A(OTA).In the absence of OTA,the fluorescence of FAM was readily quenched due to the strong binding between aptamer and PDANS.In the presence of OTA,the conformation of the OTA-specific aptamer changed from a random-coil to an antiparallel G-quadruplex,which resulted in the dissociation of the FAM from the surface of PDANS and the increasing of fluorescence intensity.This method allowed quantitative detection of OTA with a detection limit of 20 nM.Furthermore,it was applied for the detection of OTA in red wine samples and showed satisfactory results.
