GH2-34, a novel compound, inhibits Th17 differentiation and protects against Th17-mediated autoimmune diseases
【摘要】：T helper 17 cells(Th17) are a subset of pro-inflammatory T helper cells defined by their production of interleukin 17(IL-17), which are associated with autoimmune disease such as multiple sclerosis, rheumatoid arthritis, colitis and psoriasis. GH2-34 is a novel compound extracted and separated from the Myrothecium gramineum. However, there are no reports about its structure and any biological activity. Here, we showed that GH2-34 inhibited Th17 differentiation of mouse na?ve CD4 T cells in vitro. Meanwhile, GH2-34 treatment had therapeutic effect on Th17-mediated autoimmune diseases in mouse models of colitis and psoriasis, and suppressed the proportion of IL-17~+(Th17) in mice. Interestingly, GH2-34 was found to protect against sepsis and liver injury. Thus, our data demonstrates a novel compound might serve as a therapeutic agent for Th17-mediated autoimmune diseases.
